Intel opens up about its 'Knights Corner' supercomputer co-processor

Intel opens up about its Knights Corner supercomputer offering

HotChips is the show where chip makers come to show off their latest slices of silicon, and Knights Corner architect George Chrysos spilled the beans on Santa Clara's Xeon Phi co-processor. The unit's designed to bolt onto Xeon chips to help supercomputers crunch the numbers faster, by handling the "highly parallel" grunt work necessary for genetic and climate modeling, among other things. Chrysos has lofty goals for the hardware, hoping that it'll contribute to "scientific and technical progress," while we're just excited to see if it can help the company reclaim its Top 500 crown from IBM.

Tips And Strategies Associated To Internet Marketing | Make Money ...

Tips And Strategies Associated To Internet Marketing

You have to have a good Internet marketing system in place if you hope to have a successful business. If you?re not using it already, it?s time to get on board! Marketing can mean the difference between profit or failure of your business. Find your way to Internet marketing success with the ideas in the article below.

Craft your banner ads so that they stand out from the other million banner ads on the web. The more appealing and original your banner ad appears, the more people you?ll have clicking on it.

Use AJAX and Flash sparingly. Although it can help appearances on your site, it has no positive impact when it comes to increasing your search engine results. Flash should be used liberally, and additional keywords and links should be more prominent.

Increase site traffic with original content. This is even more important if your business sells the same goods as many other sites, especially if you all use identical product descriptions from the manufacturer. Write unique product descriptions incorporating keywords to make your site stand out.

If you want to improve your Internet marketing, you need to focus a lot on your emails. Remember to choose a secure e-mail service. Retaining old emails and email addresses can have a huge impact on your business, so don?t use an email service that will delete old messages. Try creating archiving arrangements for emails, particularly if you have sensitive communication with consultants and customers through email.

An FAQ page is a great way to familiarize people with your products. Offer an answer to each question, and mention products you offer as a solution to the problem when it is appropriate. Try to write in a relaxed manner, and mention your products without seeming to force them on your reader.

A successful Internet angle to follow would be to make offers to your customers for added discounts if they happen to spend over a stated amount of money. You can offer them fifteen percent off if they spend over $100. This is a motivator so people purchase more products.

A number of emotional descriptions should be included in your advertising that encourage people to become attached to your product. It will help you to become a recognized brand. Good words to use in your product descriptions include, security, energize, empower, reclaim, etc.

The advertisement of your product should always be positive and inspiring. Clearly explain how your product will help customers improve their lives or reach their goals. You want your customer to be enthusiastic about what your product can do for them, so create a level of hype that they will want to buy into.

A mailing list of your clients is essential. Give your clients the opportunity to opt-in to signing up with a subscribe link or making their email address part of the ordering process. This address can be then used for either sales letters, promotions or seeking feedback on their recent purchasing experience.

Get feedback at every stage. This is important because if you have one view of your website and your customers have a different view, your website will not be successful. Get feedback from customers, relatives and even your competition. Take all of this information under advisement and make any necessary changes.

You can use your cell phone as a work phone too, just ask your cell carrier if they can give you another number that will route to your cell phone. Be certain to acquire a dedicated phone number.

Post new content to your site every week if possible. Visitors who arrive at a site and find that the latest information available was posted months or even years ago are going to draw the wrong conclusions about your business?s current state of affairs. A site that is up-to-date and running well is inviting and encouraging for readers.

With the way the Internet operates in this modern age, you better believe that you need to employ some marketing tactics if you want to compete. You simply cannot afford NOT to use this method of advertising. If you use these tips, you will make sure your business is seen and used on the internet. This will help you succeed with any of your online endeavors

Mexico electoral judges reject challenge to Pena Nieto victory

MEXICO CITY (Reuters) - Mexico's electoral tribunal on Thursday unanimously rejected a legal bid to overturn Enrique Pena Nieto's victory in the July 1 presidential election, paving the way for the centrist to take office and press ahead with his reform agenda.

Pena Nieto, 46, and his Institutional Revolutionary Party, or PRI, were accused by runner-up Andres Manuel Lopez Obrador of laundering money and buying votes to secure victory, but the judges said there was insufficient evidence of wrongdoing.

Lopez Obrador, a leftist former mayor of Mexico City, accused the PRI of buying 5 million votes with illegal funding and plying voters with presents ranging from supermarket gift cards to fertilizer, cement and livestock.

The court must settle some smaller issues before Pena Nieto is officially declared president-elect and can take office in December, but the remaining issues are largely formalities.

"There is no proven vote buying, no evident coercion or illicit inducement," Justice Flavio Galvan said, rejecting the challenge.

Hundreds of protesters shouted their disapproval outside the tribunal's offices when the decision was announced.

Pena Nieto won the election by some 3.3 million votes, or about 6.5 percentage points. He rejected the claims of Lopez Obrador, who also unsuccessfully challenged the 2006 election result, which he lost by less than 1 percentage point.

Election experts say allegations of vote buying are nearly impossible to prove unless someone is caught in the act.

The youthful-looking Pena Nieto has had to hold back on his plans to forge deals in Congress over economic reforms due to the defiance of Lopez Obrador, who drew thousands of protesters to the streets after the July 1 election.

The protests tapped into memories of the PRI's long rule in Mexico, which lasted between 1929 and 2000 and was frequently dogged by allegations of corruption and vote-rigging.

In 2000, officials were accused of funneling more than $100 million from state oil firm Pemex to fund the PRI's unsuccessful presidential bid, when it lost to the conservative National Action Party (PAN).

Electoral authorities fined the PRI, but prosecutors failed to convict officials linked to the crime.

Lopez Obrador's allies have been accused of using similar tactics. In 2004, a close aide of Lopez Obrador was caught on camera stuffing wads of cash into a suitcase.

Once almost omnipotent in Mexico, the PRI has spent the last 12 years in opposition. Although Pena Nieto led it back to power in the July election, the party fell short of a majority in the new Congress, so it will rely on support from other parties to pass legislation.

The new Congress convenes on Saturday.

Pena Nieto has pledged to push through reforms to spur growth, such as extending the country's tax base, opening up state oil giant Pemex to more private investment, and liberalizing antiquated labor laws.

His party has also said it will seek to bring more transparency and accountability into politics in Congress.

Bait 3D: Venice Review - The Hollywood Reporter

The Bottom Line

A shark bloodbath that delivers the gore but could have used sharper teeth and a slyer sense of humor.

Venue

Venice Film Festival (Out of Competition)

Cast

Xavier Samuel, Julian McMahon, Sharni Vinson, Phoebe Tonkin

Director

Kimble Rendall

Screenwriters

Russell Mulcahy, John Kim

VENICE ? When the Australian 3D thriller Bait went into production in the wake of Snakes on a Plane, its outlandish premise earned it the informal title in industry circles of Sharks in a Mall. Combining the horror staple of a watery feeding frenzy with the timeworn ingredients of a disaster movie, this should have been B-picture bliss. But director Kimble Rendall makes the mistake of taking a silly script too seriously, and the movie never plants its tongue firmly enough in its cheek to have much fun with the mayhem.

While it?s a vast improvement on last year?s atrocious 3D Jaws wannabe, Shark Night, and less formulaic than the recent Piranha spawn, Bait is a missed opportunity. It?s also a perplexing choice for the Venice Film Festival, where only the more auteurist end of the genre spectrum usually makes an appearance. The mythic proportions of the great white shark in Australian beach culture might have been expected to yield a more distinctive bite. But the pedestrian screenplay credited to Russell Mulcahy and John Kim shows scant specificity and little feel for character development or memorable dialogue, trudging through generic themes of love, redemption, resourcefulness and survival.

STORY: Venice 2012: The Festival Gets Underway, Showing Its Feminine Side

In the pre-titles sequence, lifeguard Josh (Xavier Samuel) is engaged to marry Tina (Sharni Vinson), the sister of his buddy Rory (Richard Brancatisano). He?s preparing to give up his idyllic life in a Gold Coast surf town to relocate with her to Singapore. Still wasted from his bachelor party the night before, Josh lets Rory paddle out in his place to check on a buoy. Chomp. Josh watches helplessly as Rory gets swallowed by one of the great whites following migrating whales up the coast. Grief and guilt cause his relationship with Tina to fall apart.

One year later, dogs are howling and birds are acting crazy, which if someone were paying attention would indicate that some major natural disaster is afoot. Disillusioned Josh has taken a job at the Oceania supermarket, where Tina wanders in to shop with her new Singaporean boyfriend Steven (Qi Yuwu).

Adhering to the classic Poseidon Adventure playbook, an assortment of characters is introduced in the calm before the storm. There?s the uptight supermarket manager (Adrian Pang) who fires store clerk Ryan (Alex Russell) when his girlfriend Jaime (Phoebe Tonkin) is caught shoplifting. The cop assigned to book her is Jaime?s exasperated dad (Martin Sacks). Downstairs in the car park, a couple of blond airheads (Lincoln Lewis, Cariba Heine) make out in their vehicle, while reluctant robber Doyle (Julian McMahon) gets ready to hold up the supermarket, urged on by skuzzy hoodlum Kirby (Dan Wyllie).

STORY: Venice 2012: Inside This Year's Sober, Streamlined, Less Glitzy Festival

Rendall handles the setup confidently, but once a freak tsunami hits the town, trapping the supermarket occupants in rising water with two angry 12-foot great whites, the action becomes disappointingly routine. While there?s a welcome touch of wackiness as Steven gets suited up in makeshift armor made out of wire shelving and shopping baskets, the body count accumulates in standard fashion. Likewise the escape plans of the dwindling survivors lack imagination. Suspense is minimal.

More than the action, however, it?s the leaden dialogue that limits involvement. Samuel brings more natural charisma and conviction than the role earns, and there?s able support notably from McMahon, Tonkin, Russell and Sacks. But the uneven actors struggle to do much with their clunky lines and one-dimensional characters.

The Australian-Singaporean co-production is generally polished and effects work is solid, including the massive animatronic sharks. But at this point, the use of 3D to unleash severed body parts in underwater carnage movies is getting a little tired.

Bait?is a mistakenly earnest effort that cries out for a few more droll touches like the growling cover of ?Mack the Knife? sung on the end credits by Rendall and The Slice.

Samsung Galaxy Note II unveiled: 5.5-inch HD Super AMOLED display, Android Jelly Bean and more S Pen functionality

Samsung Galaxy Note II unveiled 55inch HD Super AMOLED display, Android Jelly Bean and more S Pen functionality

While we can't say it was a shock, Samsung's latest superphone has arrived -- and it's got a new stylus. The Galaxy Note II pushes the screen frontier to 5.5 inches wide, with another HD Super AMOLED display, this time at 1,280 x 720. Despite that expansion the phone is a mere 9.4mm thick, while it now houses a larger capacity (faster charging) 3,100mAh battery and a quad-core Exynos processor clocked at 1.6GHz. As the Galaxy Note was to the Galaxy S II, so the Note II takes some design riffs from the Galaxy S III, with the same rounded edges, glossy finish and extra software piled atop its Android base. There's also Samsung's reliable 8-megapixel camera sensor on the back, capable of 1080p video-recording.

The great news is that the Galaxy Note II will be launching on Jelly Bean -- no laborious waiting for those over-the-air updates for Google's very latest. Software additions are understandably heavily weighted towards the phablet's S Pen advances. The stylus itself now has a rubber nib, which Samsung reckons will offer an experience closer to pen and paper. User can add "Quick Commands" to their stylus gestures, while "Air View" allows you to peruse galleries and folders by floating the stylus just above the screen. Samsung's also added an Easy Clip ability to crop and share from anything beaming out from the Note II's 16:9 screen. Stylus functionality has been gifted to the calendar (S Planner) and the native email app, while the S Pen itself will now notify your phone if it's left behind. The device will launch in Titanium Grey and Marble White, arriving internationally before the end of the year. We've just managed to handle both the Galaxy Note II and that reformed stylus -- check out our hands-on here.

Manipulating the microbiome could help manage weight

Monday, August 27, 2012

Vaccines and antibiotics may someday join caloric restriction or bariatric surgery as a way to regulate weight gain, according to a new study focused on the interactions between diet, the bacteria that live in the bowel, and the immune system.

Bacteria in the intestine play a crucial role in digestion. They provide enzymes necessary for the uptake of many nutrients, synthesize certain vitamins and boost absorption of energy from food. Fifty years ago, farmers learned that by tweaking the microbial mix in their livestock with low-dose oral antibiotics, they could accelerate weight gain. More recently, scientists found that mice raised in a germ-free environment, and thus lacking gut microbes, do not put on extra weight, even on a high-fat diet.

In a study, published Aug. 26 in the journal Nature Immunology, a research team based at the University of Chicago was able to unravel some of the mechanisms that regulate this weight gain. They focused on the relationship between the immune system, gut bacteria, digestion and obesity. They showed how weight gain requires not just caloric overload but also a delicate, adjustable ? and transmissible ? interplay between intestinal microbes and the immune response.

"Diet-induced obesity depends not just on calories ingested but also on the host's microbiome," said the study's senior author Yang-Xin Fu, MD, PhD, professor of pathology at the University of Chicago Medicine. For most people, he said, "host digestion is not completely efficient, but changes in the gut flora can raise or lower digestive efficiency."

So the old adage "you are what you eat" needs to be modified, Fu suggested, to include, "as processed by the microbial community of the distal gut and as regulated by the immune system."

To measure the effects of microbes and immunity, the researchers compared normal mice with mice that have a genetic defect that renders them unable to produce lymphotoxin, a molecule that helps to regulate interactions between the immune system and bacteria in the bowel. Mice lacking lymphotoxin, they found, do not gain extra weight, even after prolonged consumption of a high-fat diet.

On a standard diet, both groups of mice maintained a steady weight. But after nine weeks on a high-fat diet, the normal mice increased their weight by one-third, most of it fat. Mice lacking lymphotoxin ate just as much, but did not gain weight.

The high-fat diet triggered changes in gut microbes for both groups. The normal mice had a substantial increase in a class of bacteria (Erysopelotrichi) previously associated with obesity and related health problems. Mice that lacked lymphotoxin were unable to clear segmented filamentous bacteria, which has previously been found to induce certain immune responses in the gut.

The role of gut microbes was confirmed when the researchers transplanted bowel contents from the study mice to normal mice raised in a germ-free environment ? and thus lacking their own microbiome. Mice who received commensal bacteria from donors that made lymphotoxin gained weight rapidly. Those that got the bacteria from mice lacking lymphotoxin gained much less weight for about three weeks, until their own intact immune system began to normalize their bacterial mix.

When housed together, the mice performed their own microbial transplants. Mice are coprophagic; they eat each other's droppings. In this way, the authors note, mice housed together "colonize one another with their own microbial communities." After weeks together, even mice with the immune defect began to gain weight. They also were able to reduce the presence of segmented filamentous bacteria in their stool.

Moving from normal chow to the high-fat diet initiated a series of related changes, the authors found. First, it altered the balance of microbes in the digestive system. These changes in the microbiome altered the immune response, which then introduced further changes to the intestinal microbial community.

These changes "provide inertia for the obese state," the authors said, facilitating more efficient use of scarce food resources.

"Our results suggest that it may be possible to learn how to regulate these microbes in ways that could help prevent diseases associated with obesity," said Vaibhav Upadhyay, first author of the study and an MD/PhD student working in Fu's laboratory. "We now think we could inhibit the negative side effects of obesity by regulating the microbiota and perhaps manipulating the immune response."

Or, 20 years from now, "when there are 10 billion people living on earth and competing for food, we may want to tilt digestive efficiency in the other direction," Fu added.

The authors cautioned, however, that with more than 500 different strains of bacteria present in the gut, "the precise microbes that promote such weight gain and the specific host responses that foster their growth need to be better established."

AP source: Redskins release K Gano, sign Cundiff

(AP) ? One day after he appeared to win the Washington Redskins kicking job, Graham Gano is out of work. His replacement is Billy Cundiff, who spent two days unemployed after getting cut by the Baltimore Ravens.

Gano was released and Cundiff signed by the Redskins on Tuesday, according to a person familiar with the situation who spoke to The Associated Press on condition of anonymity because the team had yet to make a formal announcement.

It was an abrupt development in the Redskins kicking carousel. Gano stood at this locker feeling excited and looking forward to the season Monday after his lone competition in training camp, Neil Rackers, was sent packing when the team made its first round of cuts.

But Gano's numbers have never been impressive. He has made 73.8 percent of his field goal attempts since joining the Redskins (No. 25 in the AP Pro32) late in the 2009 season, the second-worst percentage in the league over the last three seasons.

Gano, 25, missed 11 attempts in 2010, tied for most in the NFL. He had a league-high 10 misses last season, although five of those were blocked. He beat out Rackers without attempting a field goal in the Redskins' preseason games, coming out ahead based on his performance during practice.

Cundiff's statistics are only marginally better. The 32-year-old kicker has a career field goal percentage of 76.7 with the Dallas Cowboys, New Orleans Saints, Cleveland Browns and Baltimore. He joined the Ravens during the 2009 season and went to the Pro Bowl season in 2010, going 26 for 29, but last season he missed a potential game-tying, 32-yarder against the New England Patriots in the waning seconds of the AFC title game.

Cundiff also has limited range. He is 5 for 19 over his career from 50-plus yards, including just 1 for 6 last season. He was cut Sunday by the Ravens, who opted to go with rookie Justin Tucker.___

Romney's convention: Big storm, rape controversy

TAMPA, Fla. (AP) ? His Republican National Convention curtailed by a threatened hurricane, Mitt Romney conceded Sunday that fresh controversy over rape and abortion is harming his party and he accused Democrats of trying to exploit it for political gain.

"It really is sad, isn't it, with all the issues that America faces, for the Obama campaign to continue to stoop to such a low level," said Romney, struggling to sharpen the presidential election focus instead on a weak economy and 8.3 percent national unemployment.

His comments came as aides and party officials hurriedly rewrote the script for the convention, cut from four days to three because of the threat posed by approaching Tropical Storm Isaac. The storm is forecast to gain hurricane strength as it churns through the Gulf of Mexico but to pass well west of the convention city.

The revised schedule included a symbolic 10-minute session on Monday in a nearly empty hall, during which officials intend to launch a debt clock set to zero. The political objective is to show how much the government borrows throughout the convention week.

Officials did not rule out further changes because of the weather, and sidestepped when asked what might happen if, as seemed possible, the storm made landfall in the New Orleans area on the seventh anniversary of Hurricane Katrina. That storm killed 1,800 people and devastated the city.

"We're 100 percent full steam ahead on Tuesday," said Reince Priebus, chairman of the Republican National Committee, expressing confidence the one-day delay would be the extent of the cancellations.

Despite concerns about the weather, a mammoth pre-convention celebration went on as planned Sunday night, attended by thousands of delegates and others who flocked to the Rays major league baseball stadium turned into a party venue in nearby St. Petersburg.

Priebus said Romney's nomination would take place on Tuesday, as would approval of a conservative party platform.

The former Massachusetts governor delivers his acceptance speech Thursday night before a prime time TV audience, then sets out on the final leg of a quest for the presidency that spans two campaigns and more than five years.

Polls make the race a close one, with a modest advantage for President Barack Obama.

For all the Republican attempts to make the election a referendum on the incumbent's handling of the economy, other events have intervened.

An incendiary comment more than a week ago by Rep. Todd Akin, the party's candidate for a Senate seat in Missouri, is among the intrusions. In an interview, he said a woman's body has a way of preventing pregnancy in the case of a "legitimate rape." The claim is unsupported by medical evidence, and the congressman quickly apologized.

Romney and other party officials, recognizing a political threat, unsuccessfully sought to persuade Akin to quit the race. Democrats have latched onto the controversy, noting not only what Akin said but also his opposition to abortion in all cases.

"Now, Akin's choice of words isn't the real issue here. The real issue is a Republican Party ? led by Mitt Romney and Paul Ryan ? whose policies on women and their health are dangerously wrong," said a recent letter from Debbie Wasserman Schultz, the chairwoman of the Democratic Party.

The party also posted a Web video that emphasizes the Republican Party's opposition to abortion and digitally alters the Republicans' "Romney-Ryan" logo to say "Romney Ryan Akin."

Interviewed on Fox, his comments broadcast on Sunday, Romney said the controversy over Akin "hurts our party and I think is damaging to women."

Romney spent the day in New Hampshire where he has a summer home. Aides said he was spending part of his afternoon practicing his convention speech with the use of a teleprompter.

Delegates marked time as the storm raked the Florida Keys to the south of the convention city en route to a projected landfall along the Gulf Coast.

"Somebody raised the prospect of marathon Monopoly. I favor the game Risk, but we'll see," said Tom Del Beccaro, chairman of the California delegation. I think people will just be ready for Tuesday and be pretty energetic then."

Hundreds of miles away, Romney said he was concerned for the safety of those who "are going to be affected" by the storm, which is predicted to worsen into a hurricane as it heads for landfall along the Gulf Coast.

In a presidential race defined by its closeness, Republican office-holders past and present said the party must find a way to appeal to women and Hispanics, and they said the economy was the way to do it.

"We have to point out that the unemployment rate among young women is now 16 percent, that the unemployment rate among Hispanics is very high, that jobs and the economy are more important, perhaps, than maybe other issues," said Arizona Sen. John McCain, who lost to Obama in 2008.

Former Florida Gov. Jeb Bush agreed, saying that Romney "can make inroads if he focuses on how do we create a climate of job creation and economic growth." If he succeeds, "I think people will move back towards the Republican side," Bush added.

Obama leads Romney among women voters and by an overwhelming margin among Hispanics, but he trails substantially among men.

The result is a race that is unpredictably close, to be settled in a small number of battleground states.

An estimated $500 million has been spent on television commercials so far by the two candidates, their parties and supporting outside groups, nearly all of it in Florida, North Carolina, Virginia, New Hampshire, Ohio, Iowa, Colorado and Nevada. Those states account for 100 electoral votes out of the 270 needed to win the White House.

Republicans have made no secret that they are eager to expand the electoral map to include Pennsylvania, Michigan, perhaps running mate Paul Ryan's Wisconsin and even Minnesota, states with 68 electoral votes combined.

All four are usually reliably Democratic in presidential campaigns. Yet Romney has a financial advantage over the president, according to the most recent fundraising reports, and a move by the Republicans into any of them could force Obama to dip into his own campaign treasury in regions he has considered relatively safe.

Making his case for the support of female voters, Romney said in the Fox interview: "'Look, I'm the guy that was able to get health care for all of the women and men in my state. ... 'I'm very proud of what we did."

It was a rare voluntary reference to the legislation he signed as governor of Massachusetts that required the state's residents to purchase coverage, the sort of mandate that is at the heart of Obama's federal legislation that conservatives oppose and Romney has vowed to see repealed.

Romney added that the state law was put into place "without cutting Medicare, which obviously affects a lot of women."

That was a reference to the federal law, which cut more than $700 billion in projected Medicare costs to help provide health care to millions who could not otherwise afford it.

Medicare generally favors Democrats as a political issue, but Romney has aggressively sought to cut into that advantage. He released a new television ad criticizing Obama's handling of the program with a catchphrase of "It ain't right."

The streets around the convention hall were crowded with police, National Guard and other security officials, who manned checkpoints in squads rather than individually.

A few hundred protesters gathered in a park about a half-mile from the convention vowed to make their point regardless of Tropical Storm Isaac. They set out large blocks of ice spelling out the words "middle class," and left them to melt on a warm, humid day, a gesture meant to signify middle class disappearance in a tough economy.

Bush and McCain were interviewed on NBC, and Priebus spoke on CNN.

Vitamin B12 deficiency: Tracing the causes

ScienceDaily (Aug. 27, 2012) ? Vitamin B12 is vital. In collaboration with colleagues from Canada, Germany and the United States, researchers from Zurich's University Children's Hospital and the University of Zurich have succeeded in decoding a novel cause of hereditary vitamin B12 deficiency. They have discovered an important gene that determines how vitamin B12 gets into cells. Their discovery enables the diagnosis and treatment of this rare genetic disease.

Vitamin B12 is vital for cell division, the synthesis of red blood cells and the functioning of the nervous system. Unable to produce the vitamin itself, the human body has to obtain it via animal proteins. So far it has been known that on its way into the cell vitamin B12 is absorbed by little organelles, so-called lysosomes. From there, the vitamin enters the cell interior with the aid of the transport protein CblF, which was discovered by the same research team three years ago. The researchers now show that a second transport protein is actually necessary for this step, thus providing evidence of another cause of hereditary vitamin B12 deficiency.

Gene mutation prevents transport of vitamin B12

The scientists in Switzerland and Canada each examined an individual patient with symptoms of the CblF gene defect, yet without an actual defect in this gene. Using different methods, including sequencing all the coding segments of the genetic information, they were able to identify two mutations in the same gene in both patients.

The gene in question encodes the protein ABCD4, which was previously known as an ABC transporter in other cell organelles, albeit with an insufficiently defined function. It is now clear that it is a vitamin B12 transporter: "We were able to detect ABCD4 in the lysosomes of human skin cells -- right next to the already known CblF protein" explains Matthias Baumgartner, a professor of metabolic diseases at Zurich's University Children's Hospital. By adding intact ABCD4 protein to the patients' cells, the researchers were able to rescue the vitamin B12 transport and compensate for the genetic defect. "We also discovered that a targeted change in the ATP binding site of ABCD4 triggered a loss of function," says Baumgartner. Thus both ABCD4 and CblF proteins are responsible for the transfer of vitamin B12 from the lysosomes into the cell interior, and ATPase activity is involved. Baumgartner concludes: "The results obtained enable the diagnosis and treatment of this hereditary vitamin B12 deficiency."

This work was funded by the Swiss National Science Foundation (SNF) and by the Canadian Institutes of Health Research and by the Deutsche Forschungsgemeinschaft.

The Vocation Operation: Vocation Story: Continuing the Gift: Sister ...

Continuing the Gift:? Sister Madonna Joseph, O.C.D.

Sister Madonna Joseph was born in Los Angeles, California. At age nine, her family moved to the San Fernando Valley where she lived until entering Carmel.? She is the second of four children born to Marlen and Margaret Seltzer, and her home resounded with deep faith, abiding joy and laughter.? By their example, both parents taught their children about the love of God ? often using nature?s beauty to illustrate His goodness.? The family prayed the rosary and attended Mass together, and the children learned to pray morning and night prayers at an early age.

Sister Madonna Joseph?s father, a mechanic, was also an amateur ventriloquist with a fantastic sense of humor, often entertaining friends and family.? Hearts lightened when the two entered the room to entertain. Her mother, a teacher, filled the family home with the beauty of music through the many piano pieces she played ? all the way? from classical to gospel.?? Margaret Seltzer also served as a docent for the Santa Clarita Historical Society as well as for the Archives of the Archdiocese of Los Angeles.? Through these activities, she imparted her magnificent love of history to her children.? It is not surprising that the seeds of a religious vocation flourished in such a family.

During a school career day during junior high school years, the students were each asked what career they felt less inclined to choose. When it was her turn, Sister Madonna Joseph responded that her least preference would be careers in healthcare ? indeed, not to work with any type of medical equipment. God must have been smiling knowing what the future held in store for her. A graduate of Immaculate Heart College located? in Hollywood, California, she subsequently met some of the same sister-teachers who had taught her mother there as worked towards her degree in mathematics.??

Sister thought about religious life during the years after her graduation from college. However with the instability in so many religious communities during the 1970s, she decided to go into the civil service at a Veteran?s Hospital. She learned much about compassion as she observed Vietnam veterans returning home with great physical and emotional wounds from an unpopular war. Even though she enjoyed working at the Veteran?s Hospital, she felt drawn toward a deeper dimension in service to the Church, and the gentle call of God to serve Him in a religious vocation persisted. Sister Madonna Joseph remembers vividly the first time she came to Santa Teresita Hospital to ask Mother Margarita Maria for admittance into the community. She saw the sisters working together and witnessed the special presence they brought to the hospital. She knew that this was the dimension that was missing in her life.? She relates, ?Mother Margarita Maria asked if I thought I was called to the religious life. When I responded affirmatively, she said, ?Are you willing to sacrifice for Christ??? This challenge resonated in my heart.?

On September 21, 1980, Sister Madonna Joseph took up the challenge and entered Carmel. Growing up in a close-knit family where three generations had lived in the same household, respecting, appreciating and learning from another, made her transition to Carmel an easy one. She felt at home right away in Carmel where she lived with her Carmelite sisters from various generations.
????
After making her first profession of vows, she was assigned to Santa Teresita. For the next five years, she worked closely with, and learned from, Mother Margarita Maria.? Sister Madonna Joseph recalls that Mother Margarita resembled Saint Teresa of Avila with her ?holy impatience? when it came to the construction of new buildings. She remembered when she accompanied Mother Margarita Maria to one of the new building sites where the men were digging the basement. She asked them excitedly, ?When can we move in??? Sister Madonna Joseph learned from Mother Margarita Maria the intricacies of administering a healthcare facility while at the same time being interested in, and concerned for, the needs of each person.? Continuing her education, she obtained her degree in Health Care Administration.? Eight years to the day of Sister Madonna Joseph?s entrance into Carmel, God called Mother Margarita Maria home to Himself.
??
In June 2005, Sister Madonna Joseph became the Chief Executive Officer of Santa Teresita. The third Carmelite Sister to follow in the footsteps of Mother Margarita Maria, she continues today the legacy of care which has always characterized Santa Teresita.? In April 2006, Sister Madonna Joseph became the Vicar General, the assistant to Mother Regina Marie, Superior General of the Carmelite Sisters of the Most Sacred Heart of Los Angeles. Because our original sisters, now advanced in age, live at Santa Teresita, Sister Madonna Joseph relates that one of her greatest blessings is to talk to them. They tell story after story of the early days when they worked alongside our Mother Foundress and Mother Margarita Maria. She says how appreciative she is of being able to listen and learn from them about the beginnings of our community, especially the high cost of the birth of our Congregation.

When asked about her vision for Santa Teresita,
Sister Madonna Joseph answered:

?Our mission at Santa Teresita is to be ?At the Service of the Family For Life.? This is part of the legacy given to us by Mother Margarita Maria who, like our Mother Foundress, was completely at the service of her sisters, the patients, their families, our co-workers, the volunteers and the physicians. Cardinal Timothy Manning referred to Mother Margarita Maria as the woman who found the treasure in a field and sold all that she had to purchase it.? It is with this same zeal that we carry this mission into new fields of healthcare, dedicating ourselves completely to it and passing it on to future generations.?

The name ?Madonna? means ?my gift? in Italian.? For Sister Madonna Joseph, it serves as a reminder that her vocation is a gift in which she gives herself and her talents to the Lord to use as He wishes in the vineyards of Carmel.? For those of us who are privileged to know Sister Madonna Joseph, it is a reminder of the gift that she is to our Community and to those she serves. Her name is also a remembrance of the gift of a cherished legacy of loving Carmelite care at the service of the family for life ~ a gift to be passed on to generations yet to come.

MAYOR BLOOMBERG TO BAN EMPLOYEE TERMINATIONS TO CONTROL VIOLENCE IN NYC!

After today?s shooting of a former co-worker in the Empire State Building by a disgruntled fired employee, New York City Mayor Michael Bloomberg announced a new tactic to combat violence in his city. Bloomberg has long blamed violence on guns even though New York has a strict ban on gun ownership?except for liberal celebrities who own guns but call for gun bans. After the shootings today at the Empire State Building, which killed two people including the gunman, Bloomberg said:

?New York City, as you know, is the safest big city in the country and we are on pace to have a record low number of murders this year but we are not immune to the national problem of gun violence.

As you know, guns have a violent tendency to get up, leave their homes, and arbitrarily shoot people. This gun violence must stop. As such, we long ago banned gun ownership in New York City because of the violence perpetrated on innocent New Yorkers by these reprehensible, violent, unpredictable guns!

Also, we have seen that job terminations of owners of guns often causes guns to become violent, leave their homes and shoot former co-workers and by-standers. Guns tend to be very protective of their owners?especially when gun owners are terminated from their employment. Bad guns! Bad!

Since evil guns keep making their way, illegally, into New York, I am banning the terminations of any employee of any company?and especially any government worker?from being fired for any reason. This, coupled with a complete ban on guns in NYC, will eliminate all violence in New York City.?

It?s a shame a few illegal guns can cause so much danger to so many peaceful New Yorkers! As Mayor Bloomberg has taught us, people don?t kill people. It?s guns and terminations that kill people! New Yorkers support the gun bans and are well prepared to protect themselves against any violence that comes their way. New Yorkers lead the country in bagel throwing to protect themselves. They also have developed very effective techniques of begging for their lives from gunmen who don?t believe in adherence to New York?s strict gun prohibitions.

Since it appears that several innocent passers-by were injured by police gunfire today, Mayor Bloomberg is considering removing all guns from the New York City Police and arming them with three-day old bagels. He said:

?Bagels, in the hands of well-trained police officers, can be a strong deterrent to crime and violence. If you nibble away at the bagel, you can create sharp points and use it like a kung-fu throwing star like Bruce Lee. Also, bagels are environmentally friendly and will quickly decompose or be eaten by pigeons or New York?s homeless community. Believe me, no violent criminal wants to face a barrage of hardened bagels from well-trained citizens or New York?s finest!?

However, the Mayor said that New Yorkers would have to be properly licensed to own bagels older than two days. The last thing New York needs are violent criminals armed with rock-hard, sharpened bagels.

Pro-gun advocate Angry White Dude said following Bloomberg?s statement:

?Thank heavens for New York City and Mayor Bloomberg. New York is one of my favorite places in America! New York City gives home to millions of stupid idiots like Mayor Bloomberg and keeps them the hell out of Texas!?
